Description

The epitope FLDKGTYTL is derived from Epstein–Barr virus glycoprotein B (UniProt ID: P03188), encoded by the BALF4 open reading frame (ORF). The peptide corresponds to amino acids 276–284 of glycoprotein B (gB). FLDKGTYTL (epitope ID 16548) is a well-characterized HLA-A*02:01–restricted CD8⁺ T cell epitope and has been extensively studied for immune reactivity in MHC ligand assays and T cell–based functional assays.

Envelope glycoprotein B

EBV glycoprotein B is one of the five major viral envelope glycoproteins: gp350/220, gp42, BMRF2, gH/gL, and gB. Together with gH/gL, gB forms the highly conserved herpesviral fusion core. This fusion machinery is essential for viral entry, mediating the final membrane fusion step that allows delivery of the viral nucleocapsid into the host cell cytoplasm.

In epithelial cell infection, EBV attachment is initiated through integrin binding by BMRF2 and further stabilized by gH/gL. gB is subsequently recruited by the ephrin receptor tyrosine kinase A2 (EphA2), which acts as a molecular bridge bringing gB into close proximity with gH/gL. Conformational rearrangements in gB then expose conserved fusion loops, which are incorporated into the cell membrane, enabling fusion of the viral and cellular lipid membranes.
