EBV BPLF1 1395-1407 (DRA1*01:01/DRB1*03:01)
The peptide KELKRQYEKKLRQ corresponds to residues 1395–1407 of BPLF1 protein, a large tegument protein (UniProt ID: P03186) encoded by the Epstein–Barr virus (EBV, Human herpesvirus 4). This linear epitope (Epitope ID 419231) has been investigated for immune reactivity and has been experimentally validated in both T cell assays and MHC class II ligand assays, demonstrating its relevance for CD4⁺ T cell recognition.
BPLF1 is a multifunctional inner tegument protein that plays a central role during early and late stages of EBV infection. Unlike outer tegument components, BPLF1 remains at least partially capsid-associated and interacts directly with the cellular cytoskeleton. Through binding to dyneins and microtubule plus-end–associated proteins (+TIPs), BPLF1 enables active transport of the nucleocapsid toward the centrosome and subsequently to the nuclear membrane. Trafficking to the nuclear membrane is thought to resemble mechanisms described for herpes simplex virus type 1 (HSV-1) and likely involves interactions with the nuclear pore complex (NPC) mediated by BPLF1.
BPLF1 is an integral component of the capsid-associated tegument complex (CATC), together with BGLF1 and BVRF1. BPLF1 is a known caspase-1 substrate, and its N-terminal fragment functions as a highly active deubiquitinase, modulating host protein degradation pathways and deregulating signaling cascades such as mTOR. In contrast, the C-terminal fragment of BPLF1 is thought to be incorporated into the CATC, contributing to capsid stabilization by binding to penton structures during late stages of assembly.
